We have users with two email addresses (custom field "email 2").
Is it possible that an e-mail is sent to both the email addresses when an new ticket is posted or when a reply to a old ticket is being made?

No. You'd have to fork (create a copy of) the plugin that send the emails and customize it. That's a bad idea since it will no longer receive updates from us.
