Description of my issue:

I've created a backup of a site and installed it on a sub-domain for testing purposes. (To check extension and Joomla updates before installing those on live sites) Is it possible to synchronize the testdomain with the live site, so that changes made to the live site are the same on the test site, without having to make a complete new backup file? I don't mind that changes made on the test site are overwritten.

No, you can't. You'd need to take a new backup and restore it on the test site.
