Hello, it's possible to put in automatic offline site while Akeeba Pro backup site and then when finish backup put site online in automatic ?

It is not possible to do that automatically. You can set the site offline in the Global Settings screen, then turn it back on after the backup completes. While there is a small risk of data corruption caused by data posted during the backup, it is a pretty small risk for most sites. It is usually more trouble than it's worth to take the site offline.
