Description of my issue: I use S3 as my backup location, but am always paranoid about losing data. Is there any way to back up to S3 and one other location, say Dropbox or SugarSync?

No, there is no way to do that in a single backup. You could set up different backup profiles and back up to different locations, either do two backups per night or alternate profiles to get them to multiple locations.
